Alexandre la conquit avec beaucoup de difficultés entre 329 et 327 avant notre ère, en grande partie avec l'aide de forces auxiliaires locales. Pendant son séjour, Alexandre épousa une Bactriane, Roxanne, afin qu'elle l'aide à contrôler la région. Après la mort d'Alexandre, la Bactriane faisait partie de l'[empire séleucide](https://www.worldhistory.org/trans/fr/1-246/empire-seleucide/). Les nombreuses difficultés rencontrées par les rois séleucides et les attaques de Ptolémée II d'Égypte donnèrent à Diodote, satrape de Bactriane, l'occasion de déclarer son indépendance (c. 255 av.
